Unmodified Single-Stranded DNA

DNA oligonucleotides are short, synthetic sequences—single or double-stranded—used in a wide range of molecular biology applications, including PCR, sequencing, gene editing, and hybridization assays.

Our Oligo Entry tool enables precise sequence design and supports an extensive range of chemical modifications to meet diverse experimental requirements.

Each oligo is delivered fully deprotected and desalted, removing residual small molecules and synthesis by-products. Final products undergo quality verification using our proprietary electrospray ionization mass spectrometry (ESI-MS*) to confirm molecular integrity and are quantified in duplicate by UV spectrophotometry for accurate yield reporting.

*Note: ESI-MS analysis is not performed on oligos containing mixed bases due to sequence ambiguity.

Technical Notes

Every DNA Oligo you receive will be deprotected and desalted to remove small molecule impurities. In addition, your oligos will be quality controlled via proprietary ESI-MS*, and quantified twice by UV spectrophotometry to provide yield measurements.

* With the exception of mixed base oligos, which could potentially represent multiple sequences and therefore cannot be evaluated by ESI -MS.

For high-precision applications such as multiplex PCR, cloning, mutagenesis, or antisense/RNAi experiments, enhanced purification is often required. All purified oligos up to 100 bases with purification are QC-verified by gel electrophoresis or LC-MS and backed by a purity guarantee.

Pricing is per oligo for purification only and does not include synthesis cost. All In vivo DNA options (In vivo GF and In vivo HPLC) include 0.22 μm sterile filtration and Na⁺ counterion exchange (final sodium salt form) in a low-endotoxin, nuclease-controlled workflow.

Technical Notes

Purification:
All Bio-Synthesis' oligos include desalting and deprotection at no additional charge. We strongly recommend oligonucleotide longer than 50 bases to be purified by poly acrylamide gel electrophoresis (PAGE). If you are unsure about the purification level you need for your research, see our technical bulletin titled What type of purification I need for my applications or contact us for assistance.

Minimum yield
Yield will vary based upon base composition, purification, oligo length, scale and modifications. That's why we have a set guaranteed quantity.

Guaranteed Quality
All custom oligo synthesis are routinely checked for length, purity, and composition. Long oligos are typically quality checked by PAGE, ESI-MS, and/or CE. Our global quality control capabilities include MALDI-TOF mass spectrometry, electrospray ionization mass spectrometry (ESI-MS), capillary electrophoresis (CE), and polyacrylamide gel electrophoresis (PAGE). If you are dissatisfied and report an oligo problem within 30 days of order receipt, we will gladly remake your oligo free of charge or provide a credit for the purchase amount. All orders are shipped with a Quality Assurance document that includes measured yields, melting temperature, MW and µg/OD with every oligo.

FAQ

What purification do I need?

For routine PCR, Desalt is usually sufficient. For long (=100 nt), modified, or probe-grade oligos, choose HPLC or PAGE. See Purification for guidance.

How long can you synthesize?

Standard DNA up to ~200 nt with long-oligo protocols. For longer constructs, we recommend assembly from overlapping oligos.

Do you support GLP/cGMP production?

Yes. We support RUO, GLP, and cGMP with appropriate QA oversight and documentation.

Can I get oligos in plates with barcodes?

Yes—96/384‑well plates, lyophilized or in buffer, with barcoded labels and downloadable plate maps.

What QC is included?

MS identity and analytical HPLC purity (per selected grade). Optional endotoxin/bioburden for regulated programs.
