Why do PTM peptides often fail at other suppliers?
PTM peptides frequently fail due to on-resin aggregation, incomplete modification, or PTM instability. We mitigate this using orthogonal protection, stepwise validation, and purification methods tailored to each PTM class (e.g., phospho, sulfated, acetylated peptides).
Is lipidation just attaching a fatty acid?
No. Lipidation is an architecture-level modification. The lipid type, attachment site (N-terminus vs side-chain), linker choice, and sequence hydrophobicity all determine solubility, aggregation risk, and biological performance. We design lipidated peptides accordingly.
How do you ensure site-specific modification?
Site specificity is achieved through single-handle design, residue-selective chemistry (e.g., cysteine-only), and protection strategies that prevent off-target labeling—critical for conjugates and quantitative studies.
Do modifications affect MS confirmation?
Yes. Many modifications alter ionization or fragmentation. We adjust MS methods accordingly and report monoisotopic or exact mass confirmation whenever applicable.
